Summary

Autoimmunity is the failure of an organism to recognize its own constituent parts as self, which allows an immune response against its own cells and tissues. Any disease that results from such an aberrant immune response is termed an Autoimmune Disease.

Antibodies against dsDNA belong to the group of Anti Nuclear Antibodies(ANA) which appear in several rheumatoid diseases, in particular presence of autoantibodies to native double-stranded DNA is typical for the clinical picture of Systemic Lupus Erytrematosus (SLE).

Antibodies against dsDNA appear in the active phase of the SLE and their concentration is prognostic to the severity of disease. DsDNA autoantibodies quantification become really useful in the follow up of the therapy.

Anti-dsDNA are present in approximately 90% of SLE patients.

Test Principle

Microplates are coated with a dsDNA (Plasmid Rec). In the 1st incubation, the solid phase is treated with diluted samples and anti dsDNA IgG are captured, if present, by the solid phase.

After washing out all the other components of the sample, in the 2nd incubation bound anti dsDNA IgG are detected by the addition of anti hIgG antibody, labelled with peroxidase (HRP).

The enzyme captured on the solid phase, acting on the substrate/chromogen mixture, generates an optical signal that is proportional to the amount of anti dsDNA IgG antibodies present in the sample. IgG in the sample may be quantitated by means of a standard curve calibrated on WHO standard preparation for anti-dsDNA antibodies (Wo/80).
