α2-macroglobulin
Human a2-macroglobulin (AMG) is a plasma glycoprotein of molecular weight 725,000, containing four identical subunits bound together by -S-S- bonds. This protein binds to a wide variety of proteases, usually resulting in inhibition of the activity of these proteases. Increased levels are reported in nephrotic syndrome, pregnancy, liver diseases, diabetes mellitus, inflammatory diseases, bronchopneumonia, congenital cardiac disease. Decreased levels are reported in fibrinolysis, acute pancreatitis, biliary or renal stones, liver tumors, gastroduodenal ulcers, myocardial infarction.
Immunonephelometry is applied. This method involves measuring the light scattered by insoluble complexes formed by reaction between specific protein in samples and its respective antiserum, and the amount of scattered light is directly proportional to the concentration of the protein under condition that antiserum is in excess. Concentrations are automatically calculated by reference to a calibration curve stored in the instrument.
